NMN
The biomarker moves. The outcomes have not.
NMN (nicotinamide mononucleotide) is a precursor to NAD+, a molecule central to cellular energy metabolism that declines with age. We grade NMN C+ on evidence and D+ for worth-it at about $30-60 per month.

What it does
NMN (nicotinamide mononucleotide) is a precursor to NAD+, a molecule central to cellular energy metabolism that declines with age. Raising NAD+ became one of longevity's biggest bets.
What the research says
Reliably raises blood NAD+ levels
Evidence: strong. Jan 2026 RCT (n=65): 1000 mg/day roughly doubled blood NAD+ in 14 days, comparable to NR. UTokyo trial (n=62, 65+, 1250 mg/day, 24 weeks, NPJ Aging): sustained NAD+ elevation across the trial.
May improve muscle insulin sensitivity in prediabetes
Evidence: emerging. 2022 trial: 250 mg/day for 10 weeks improved clamp-measured muscle insulin sensitivity 25% in overweight prediabetic postmenopausal women. Not replicated broadly.
No demonstrated effect on hard health outcomes yet
Evidence: moderate. UTokyo 24-week trial: no significant effect on hearing (primary), physical function, or other health outcomes despite NAD+ rise. 2026 meta-analysis (15 RCTs): safe and well tolerated, no broad metabolic benefits; slight diastolic BP dip and HOMA-IR trend only.
DRAFT: biomarker case (NAD+ elevation) is now strong across multiple RCTs, but the UTokyo 24-week trial found no health-outcome effects and the 2026 meta-analysis found no broad metabolic benefits. Biomarker strong, outcomes thin.
What the experts say
Every position links to its source. We include skeptics alongside supporters.
David Sinclair
Source, 2026-08-27 Takes 1 gram of NMN every morning, as stated in his book Lifespan.
Charles Brenner
“NMN doesn't get into cells as NMN. it gets in as NR and there's a safe source of NR so why are we still talking about NMN?”
Source, 2023-10-17 Argues NMN has far less human research and no known safe source, and that NMN enters cells as NR anyway, so NR is the better-supported choice.
Andrew Huberman
“I don't expect to live longer because of taking NAD. I just sort of like the effect that it appears to give me.”
Source, 2026-09-26 Takes NMN powder for a perceived energy effect but doubts it extends lifespan.
Bryan Johnson
Source, 2026-03-24 Takes 500 mg of NMN or NR six days a week to boost NAD+ for energy, DNA repair, and cellular health.
Is it worth it?
DRAFT: you are paying $40-80/month mostly for a biomarker increase with no proven outcome benefit yet.
Typical cost
$30-60/month
Common form, not a brand recommendation
Dosage contextemerging
250 to 500 mg per day in published trials; no established long-term dosing.
Safety notes
Short-term trials report good tolerability, but long-term human safety data does not exist. Regulatory status has shifted over time; check current rules in your country. Talk to your doctor. This page is information, not medical advice.
